Properties & Usage

Unit Definition

One unit is defined as the amount of enzyme required to protect 1 µg (dam-) Lambda DNA in 1 hour at 37°C in a total reaction volume of 10 µl against cleavage by MboI restriction endonuclease.

Supplement with 640 µM S-adenosylmethionine (SAM)
Incubate at 37°C

Control Protection Assay

One unit of dam Methyltransferase is incubated with 1 µg of (dam-) Lambda DNA in 10 µl 1X dam Methyltransferase Reaction Buffer, supplemented with 640 µM S-adenosylmethionine, for one hour at 37°C followed by 15 minutes inactivation at 65°C. The extent of protection by dam Methyltransferase is determined by the addition of 10 units of MboI restriction endonuclease in 40 µl 1X NEBuffer 3 supplemented with 10 mM MgCl2 followed by incubation at 37°C for one hour. Results are analyzed on agarose gel.

Tech Tips

Always add the SAM to the reaction buffer just before doing the methylation reaction, because the SAM is unstable in an aqueous solution.

Keep the SAM frozen at -20C for a longer shelf life, fresh SAM is important for optimal methylation reactions.
