Description

9°N™ Reverse Gyrase is a thermostable Type IA topoisomerase that introduces positive supercoils into double-stranded closed-circular DNA in the presence of ATP (1).

Product Source

An E. coli strain that carries a cloned gene from a thermophilic marine archaea Thermococcus sp. (strain 9°N) (2) with a C-terminal His-tag.

Properties & Usage

Unit Definition

One unit is defined as the amount of enzyme required to supercoil 0.5 µg of relaxed pUC19 plasmid in 15 minutes at 80°C, in a total reaction volume of 40 µl.

Product Notes

  1. The reaction requires ATP and Mg2+ (3, 4). The supplied Gyrase Reaction Buffer incorporates these essential components within an optimized formulation, thereby facilitating enhanced gyrase activity.
  2. In the absence of ATP, the enzyme will nick circular DNA (5).
